Common medium and basic characteristics of cell culture

1, RPMI-1640 Medium
RPMI-1640 is widely used in mammals, special hematopoietic cells, normal or malignant hyperplasia leukocytes, and hybridoma cell culture is a widely used medium. Mainly used for suspension cell culture. Other lymphocytes, T-cell lymphoma cells, and HCT-15 epithelial cells such as K-562, HL-60, Jurkat, Daudi, and IM-9 can be used as reference.
2, Minimum Essential Medium (MEM)
Also known as the minimum essential medium, it contains only 12 essential amino acids, glutamine and 8 vitamins. The composition is simple and can be widely adapted to the cultivation of mammalian cell types in various established cell lines and different places. MEM-Alpha is generally used to culture some cell types that are difficult to culture, while other cell lines that have no special features can be cultured in almost MEM.
3, DMEM-high sugar (standard type)
It is a widely used medium that can be used in many mammalian cell cultures and is more suitable for high-density suspension cell culture. It is suitable for clonal culture with poor adhesion, but does not want it to leave the original growth point. It can also be used for the culture of myeloma cells and DNA transfected transformed cells in hybridomas.
4, DMEM-low sugar (standard type)
It is a widely used medium that can be used in many mammalian cell cultures. Low glucose is suitable for dependent adherent cell culture, and is particularly suitable for tumor cell culture with fast growth rate and poor adhesion.
5, DMEM / F12
DMEM/F12 medium is suitable for culture of clonal density. The F12 medium is complex in composition and contains a variety of trace elements. It is combined with DMEM in a ratio of 1:1 and is called DMEM/F12 medium (DME/F12medium). It is used as a basis for developing serum-free formulas to utilize F12 to contain rich components and DMEM has the advantage of containing a higher concentration of nutrients. This medium is suitable for mammalian cell culture under conditions of low serum levels. One of the improvements to enhance the buffering capacity of the medium was to add 15 mM HEPES buffer to DMEM/F12 (1:1).
6, McCoy's 5A
Designed primarily for the culture of sarcoma cells, McCoy's 5A Medium supports the growth of a variety of primary grafts (such as bone marrow, skin, lungs, and spleen), except for general primary cell culture. Tissue biopsy culture, some lymphocyte cultures, and growth support for some difficult-to-cultivate cells. For example, Jensen rat sarcoma fibroblasts, human lymphocytes, HT-29, BHL-100 and other epithelial cells.
7. Iscove's Modified Dulbecco Medium (IMDM)
Guilber and Iscove modified Dulbecco'Medium to Iscove's Medium for culturing red blood cells and macrophage precursors. This culture contains selenium, additional amino acids and vitamins, sodium pyruvate and HEPES. And replaced the ferric nitrate with potassium nitrate. IMDM is also capable of promoting the growth of mouse B lymphocytes, LPS-stimulated B cells, bone marrow hematopoietic cells, T cells and lymphoma cells. IMDM is a nutrient-rich medium, so it can be used for rapid proliferation of high-density cells.
8, M-199 Medium
In 1950, Morgan successfully developed a cell culture medium with a defined chemical composition, M-199, which was mainly used for chicken embryo fibroblast culture. This culture must be supplemented with serum to support long-term culture. M-199 can be used to culture cells of various species and to culture transfected cells.
9, Leibovitz Medium (L-15)
The L-15 medium is suitable for the culture of rapidly proliferating tumor cells for culturing tumor cell lines in the absence of CO2. This culture solution uses a phosphate buffer system, and the amino acid composition is further improved, and glucose is replaced by galactose.
10. Ham's F-10 medium: adapted to the culture of mouse cells and human diploids.
11. Ham's F-12 medium: It can be applied with little serum addition, especially suitable for single cell culture and cloning culture. It is a basic culture medium commonly used in serum-free culture.
12. William's Medium E: Long-term cell culture for rat liver epithelial cells.
13. MCDB 131 medium: used to culture endothelial cells.
14, Opti-MEM I Reduced Serum Media: used to culture hematopoietic cells.
15. Phytohemagglutinin (PHA): Increases cell transformation and DNA synthesis.
